Cependant, la transformation de la lignocellulose repr\u00e9sente un enjeu de taille. En particulier, la valorisation de la lignine, polym\u00e8re aromatique, pr\u00e9sente un fort potentiel mais reste un point d\u2019achoppement important en raison de la difficult\u00e9 de sa d\u00e9construction.<\/p><p>La nature regorge pourtant d\u2019organismes pour lesquels la biomasse lignocellulosique est \u00e0 la base du r\u00e9gime alimentaire. C\u2019est notamment le cas des insectes xylophages comme les termites dont les communaut\u00e9s microbiennes endosymbiotiques sont capable de convertir la lignocellulose en ac\u00e9tate utilis\u00e9 par l\u2019h\u00f4te comme source d\u2019\u00e9nergie. En revanche, notre connaissance actuelle de la d\u00e9gradation de la lignine par des communaut\u00e9s microbiennes issues de termites reste insuffisante. Nous nous proposons donc, dans cette th\u00e8se, d\u2019\u00e9tudier des consortia microbiens issus de termite et leur implication potentielle dans la d\u00e9lignification.<\/p><p>Dans le cadre du projet europ\u00e9en ZELCOR (\u201cZero Waste Ligno-Cellulosic Biorefineries by Integrated Lignin Valorisation\u201d) fond\u00e9 par le partenariat public-priv\u00e9 BBI (Bio-Based Industries), nous avons proc\u00e9d\u00e9 \u00e0 l\u2019enrichissement en batchs successifs de consortia microbiens sur diff\u00e9rents substrats et dans diff\u00e9rentes conditions afin d\u2019obtenir un consortium capable de d\u00e9grader la lignine puis l\u2019avons cultiv\u00e9 en bior\u00e9acteurs. Afin de montrer le potentiel de consortia microbiens issus de termite pour la valorisation de la lignine, nous avons coupl\u00e9 la caract\u00e9risation structurelle des substrats utilis\u00e9s au cours de la digestion par hydrolyse acide, 2D HSQC NMR et\u00a0<span style=\"font-size: 9.75px; line-height: 0; position: relative; vertical-align: baseline; top: -0.5em;\">13<\/span>C-IS py-GC-MS \u00e0 la caract\u00e9risation des communaut\u00e9s microbiennes par m\u00e9tag\u00e9nomique \u00ab\u00a0shotgun\u00a0\u00bb.<\/p><p>Cette approche coupl\u00e9e nous a permis de d\u00e9montrer le potentiel lignolytique, aussi bien en milieu ana\u00e9obie qu\u2019a\u00e9robie de communaut\u00e9s microbiennes issues de termites. Nous avons \u00e9galement pu enrichir et \u00e9tudier un consortium lignocellulolytique.\u00a0 Gr\u00e2ce \u00e0 la reconstruction de MAGs, nous avons pu identifier des bact\u00e9ries et enzymes d\u2019int\u00e9r\u00eat biotechnologique.
