Ce r\u00e9sultat a montr\u00e9 que toutes les mol\u00e9cules non fonctionnelles d&rsquo;ARNm ne sont pas d\u00e9truites, r\u00e9v\u00e9lant le gaspillage d&rsquo;une partie des ressources intracellulaires et apportant un nouvel \u00e9clairage sur le contr\u00f4le qualit\u00e9 des ARNm chez les bact\u00e9ries.<\/strong><\/p>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-54aa915 elementor-widget elementor-widget-text-editor\" data-id=\"54aa915\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t\t\t\t\t\t<p>mRNA is a central molecule in gene expression. Produced by gene transcription, it is the molecular template for transforming genetic information into protein through translation. mRNA is a highly unstable molecule, rapidly degraded by a series of RNases present in the cell.\u00a0<\/p>\n<p>The team has developed a strategy for measuring stability along the entire length of the mRNA molecule. Using a set of specific probes, it is now possible to determine the stability parameters and local concentrations of different portions of a molecule, to determine whether a molecule is homogeneously degraded or whether certain portions are more stable (or less stable) than others.<\/p>\n<p>Cette cartographie a permis de montrer, chez la bact\u00e9rie Escherichia coli, que lorsque l\u2019\u00e9longation de la traduction est stopp\u00e9e pr\u00e9matur\u00e9ment et qu\u2019une partie de la mol\u00e9cule n\u2019est plus couverte par les ribosomes, l\u2019ARNm est d\u00e9stabilis\u00e9 de fa\u00e7on homog\u00e8ne. Cette d\u00e9stabilisation est principalement due \u00e0 une enzyme (RNase E) et \u00e0 sa capacit\u00e9 \u00e0 former un complexe avec d\u2019autres prot\u00e9ines (d\u00e9gradosome). En revanche, nous avons d\u00e9couvert une h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 au niveau des mol\u00e9cules d\u2019ARNm qui peuvent pr\u00e9senter des longueurs diff\u00e9rentes dans la cellule. Ce r\u00e9sultat a r\u00e9v\u00e9l\u00e9 que toutes les mol\u00e9cules non fonctionnelles ne sont pas d\u00e9truites, apportant ainsi un nouvel \u00e9clairage sur le contr\u00f4le qualit\u00e9 des ARNm chez les bact\u00e9ries qui n&rsquo;aboutit pas \u00e0 la d\u00e9gradation totale de ces mol\u00e9cules.<\/p>\n<p>The quality control of mRNAs contributes to the proper functioning of the cell by avoiding the expression of unnecessary functions that waste intracellular resources. The TBI team is now focusing its research on characterizing the players involved in mRNA quality control mechanisms, with the aim of gaining a better understanding of this molecular heterogeneity and, ultimately, better controlling it. Controlling mRNA degradation is a lever for optimizing the expression of genes of interest.
