Etienne\u00a0SEVERAC<\/td><td>INSA &#8211; Toulouse<\/td><td>Co-encadrant de th\u00e8se<\/td><\/tr><\/tbody><\/table><p>\u00a0<\/p><p>\u00a0<\/p><table border=\"0\" width=\"98%\"><tbody><tr><td><b>R\u00e9sum\u00e9\u00a0:<\/b><\/td><\/tr><tr><td><div>Pour faire face aux impacts de la chimie conventionnelle et des produits p\u00e9trosourc\u00e9s sur la sant\u00e9 humaine et les \u00e9cosyst\u00e8mes, la chimie verte et les macromol\u00e9cules d&#8217;origine biologique sont une source infinie d&#8217;innovation. Ce projet s\u2019inscrit dans une approche de bioinspiration et repose sur l\u2019utilisation d\u2019enzymes bact\u00e9riennes dans l\u2019objectif d\u2019\u00e9valuer leur potentiel pour : (1) la glucosylation de mol\u00e9cules marines pour la cr\u00e9ation de filtres solaires, et (2) l&#8217;assemblage de polysaccharides marins et bact\u00e9riens r\u00e9ticul\u00e9s par des prot\u00e9ines \u00ab sur mesure \u00bb. Dans le premier cas, il s\u2019agit de produire des mol\u00e9cules inspir\u00e9es des propri\u00e9t\u00e9s anti-UV retrouv\u00e9es chez certains compos\u00e9s du mucus et des yeux des poissons. Dans le second, d\u2019une strat\u00e9gie inspir\u00e9e des associations entre polysaccharides et prot\u00e9ines que l\u2019on trouve dans les peptidoglycanes bact\u00e9riens ou les carapaces des crustac\u00e9s. Pour le premier projet, les mol\u00e9cules cibles sont les mycosporines, des m\u00e9tabolites secondaires pr\u00e9sents dans une grande vari\u00e9t\u00e9 d&#8217;organismes et reconnues pour leur forte capacit\u00e9 d\u2019absorption des rayons UV et leur pouvoir antioxydant. En prenant la mycosporine-s\u00e9rinol (MSer(OH)) comme mod\u00e8le, une vingtaine d\u2019\u03b1-transglucosylases de la famille GH70 qui utilisent le saccharose comme substrat donneur d\u2019unit\u00e9s glucosyle ont \u00e9t\u00e9 test\u00e9es. Une d\u2019entre elle s\u2019est particuli\u00e8rement d\u00e9marqu\u00e9e, glucosylant la mycosporine avec un taux de conversion de 95%. Les caract\u00e9risations en RMN et spectrom\u00e9trie de masse ont montr\u00e9 l&#8217;ajout de 1 \u00e0 3 unit\u00e9s glucosyle, l\u2019esp\u00e8ce pr\u00e9dominante ayant 2 unit\u00e9s sucre sur le m\u00eame carbone. Dans un deuxi\u00e8me temps, une cascade enzymatique a \u00e9t\u00e9 d\u00e9velopp\u00e9e afin d&#8217;allonger la cha\u00eene glucidique pr\u00e9alablement obtenue, toujours \u00e0 partir de saccharose et de ces m\u00eames enzymes. Une large gamme de MSer(OH) glucosyl\u00e9es a ainsi \u00e9t\u00e9 g\u00e9n\u00e9r\u00e9e, variant par la nature des liaisons osidiques et\/ou la taille de la chaine (10 &lt; DP &lt; ~750 000). La photostabilit\u00e9 et les capacit\u00e9s antioxydantes des produits obtenus sont \u00e9quivalentes \u00e0 celles de la MSer(OH) libre ou d\u2019antioxydants reconnus, et peuvent donc rivaliser avec des filtres solaires commerciaux. Notamment, une MSer(OH) greff\u00e9e sur une cha\u00eene de dextrane de plus de 108 g\/mol pr\u00e9sente des propri\u00e9t\u00e9s texturantes int\u00e9ressantes, la rendant d\u2019autant plus prometteuse pour des formulations m\u00e9dicales ou cosm\u00e9tiques. Dans le deuxi\u00e8me projet, le choix s\u2019est port\u00e9 sur le dextrane, la chitine, le chitosane et l\u2019agarose car ce sont des polysaccharides largement disponibles et d\u00e9j\u00e0 reconnus dans le domaine des biomat\u00e9riaux, notamment pour des applications m\u00e9dicales. L\u2019assemblage physique de ces biopolym\u00e8res est bas\u00e9 sur la cr\u00e9ation de prot\u00e9ines r\u00e9ticulantes sur-mesure nomm\u00e9es &#8220;bridges&#8221;, correspondant \u00e0 l\u2019association de 2 modules de liaison aux glucides (CBM) &#8211; issus d\u2019enzymes d\u00e9gradant ou synth\u00e9tisant ces polym\u00e8res &#8211; li\u00e9s entre eux par une s\u00e9quence peptidique plus ou moins flexible. Deux biblioth\u00e8ques de quatre bridges ont \u00e9t\u00e9 con\u00e7ues, produites de mani\u00e8re recombinante chez E. coli, purifi\u00e9es, et leur affinit\u00e9 pour leurs biopolym\u00e8res respectifs confirm\u00e9e. Ensuite, des premiers hydrogels physiques ont \u00e9t\u00e9 d\u00e9velopp\u00e9s en assemblant les polysaccharides avec les \u00ab bridges \u00bb, menant \u00e0 des diff\u00e9rences de texture et de viscosit\u00e9. Des analyses de rh\u00e9ologie et de microscopie ont confirm\u00e9 que ces biomat\u00e9riaux sont dynamiques et rh\u00e9ofluidifiants, donc tr\u00e8s int\u00e9ressants comme hydrogels injectables pour l\u2019administration de m\u00e9dicament. Dans l\u2019ensemble, ces premi\u00e8res preuves de concept sont prometteuses et montrent le potentiel de ces mat\u00e9riaux bioinspir\u00e9s pour diff\u00e9rentes applications. Les plus int\u00e9ressantes sont celles envisag\u00e9es dans le domaine de la sant\u00e9, car elles permettraient la formulation de produit respectueux \u00e0 la fois de la sant\u00e9 humaine et des
