In many Gram-negative bacteria, RNA degradosomes are proteins located at the inner membrane of the bacterium. The reason for this localisation was not understood.<br>Using mutants that have free degradosomes in the cytoplasm, the scientists, in a paper published in the journal<em>\u00a0Plos Biology<\/em>the results of this study show that this localisation is important to preserve the quality of ribosome assembly.<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Read this article on <a href=\"https:\/\/www.insb.cnrs.fr\/fr\/cnrsinfo\/controle-qualite-de-lassemblage-des-ribosomes-bacteriens-par-le-degradosome-darn\" target=\"_blank\" rel=\"noreferrer noopener\">le site de l&rsquo;INSB.<\/a><\/p>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<p class=\"wp-block-paragraph\"><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">La maturation et la d\u00e9gradation des ARN fa\u00e7onnent le transcriptome en g\u00e9n\u00e9rant des mol\u00e9cules stables n\u00e9cessaires \u00e0 la traduction (ARNr et ARNt) et en facilitant le turnover des ARNm n\u00e9cessaire au contr\u00f4le post-transcriptionnel de l&rsquo;expression des g\u00e8nes. Chez les bact\u00e9ries et dans le chloroplaste des plantes, les d\u00e9gradosomes d\u2019ARN sont des complexes multi-enzymes qui maturent et d\u00e9gradent les ARN. Les d\u00e9gradosomes d\u2019ARN contenant la RNase E sont attach\u00e9s \u00e0 la membrane cytoplasmique interne chez une large famille de bact\u00e9ries Gram-n\u00e9gatives (\u03b2- et \u03b3-prot\u00e9obact\u00e9ries). Jusqu&rsquo;\u00e0 pr\u00e9sent, la raison de leur localisation \u00e0 la membrane n&rsquo;\u00e9tait pas comprise.<\/p>\n\n\n\n<p class=\"wp-block-paragraph translation-block\">In this study, scientists show that a mutant strain of Escherichia coli, in which the RNA degradosome is located inside the cell, accumulates 20S and 40S particles that are precursors of defective ribosome assembly. Their protein composition is abnormal and they contain precursors of RNase E-cleaved rRNAs. They concluded that the RNA degradosomes located inside the cell in the mutant strain interfere with the assembly of ribosomal subunits.<\/p>\n\n\n\n<p class=\"wp-block-paragraph translation-block\">To better understand the mechanism, they showed in vitro that rRNAs contained in intact ribosomes are resistant to cleavage by RNase E, whereas protein-free rRNAs are easily degraded. This demonstrates that the compact structure of mature ribosomes protects rRNAs from degradation by RNase E.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">La plupart des op\u00e9rons des ARNr sont \u00e0 proximit\u00e9 dans le nucl\u00e9o\u00efde, sugg\u00e9rant l&rsquo;existence d&rsquo;un nucl\u00e9ole bact\u00e9rien. Les premi\u00e8res \u00e9tapes de l&rsquo;assemblage des ribosomes bact\u00e9riens impliquent le repliement co-transcriptionnel des ARNr et la liaison des prot\u00e9ines ribosomales (r-prot\u00e9ines) dans le nucl\u00e9o\u00efde. De multiples voies d&rsquo;assemblage agissent en parall\u00e8le dans des blocs coop\u00e9ratifs de repliement des ARNr pour constituer les domaines structurels des sous-unit\u00e9s ribosomales matures. \u00c9tant donn\u00e9 la complexit\u00e9 du processus d&rsquo;assemblage des ribosomes, il n&rsquo;est pas surprenant que des interm\u00e9diaires d\u00e9fectueux apparaissent occasionnellement.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Les d\u00e9gradosomes d\u2019ARN fix\u00e9s \u00e0 la membrane dans les cellules de type sauvage contr\u00f4leraient la qualit\u00e9 de l&rsquo;assemblage des ribosomes apr\u00e8s la lib\u00e9ration des interm\u00e9diaires du nucl\u00e9o\u00efde. L&rsquo;attachement du d\u00e9gradosome d\u2019ARN \u00e0 la membrane cytoplasmique interne des bact\u00e9ries emp\u00eache la d\u00e9gradation inutile des pr\u00e9curseurs d&rsquo;ARNr, expliquant ainsi pourquoi cet attachement est conserv\u00e9 dans toutes les \u03b2- et \u03b3-prot\u00e9obact\u00e9ries.
