En \u00e9tudiant le passage d\u2019une croissance sur glucose \u00e0 une croissance sur xylose, l\u2019\u00e9quipe BLADE a pu montrer que la dur\u00e9e de la latence est en fait due \u00e0 l\u2019\u00e9mergence d&#8217;une sous-population de cellules adapt\u00e9es qui va ensuite devenir la population principale. En identifiant le m\u00e9canisme mol\u00e9culaire contr\u00f4lant l&#8217;\u00e9mergence de la sous-population, les chercheurs ont pu allonger ou r\u00e9duire la dur\u00e9e de cette phase d\u2019adaptation. Ce travail est d&#8217;une importance particuli\u00e8re car il d\u00e9montre la complexit\u00e9 inattendue des populations dites \u00ab\u00a0pures\u00a0\u00bb au cours de la croissance sur des substrats mixtes et ouvre la voie vers de nouvelles strat\u00e9gies pour une meilleure utilisation des sources de carbone durables en bio\u00e9conomie.<\/strong><\/p><p>La transition m\u00e9tabolique glucose-xylose pr\u00e9sente un int\u00e9r\u00eat croissant comme mod\u00e8le pour explorer l&#8217;adaptation cellulaire puisque ces mol\u00e9cules sont les principaux substrats r\u00e9sultant de la d\u00e9construction de la biomasse lignocellulosique. Si la bact\u00e9rie <em>Escherichia coli<\/em> est capable de rapidement consommer le glucose, il n\u2019en va pas forc\u00e9ment de m\u00eame pour sa capacit\u00e9 \u00e0 basculer sur l\u2019utilisation du xylose lorsque le glucose a \u00e9t\u00e9 enti\u00e8rement utilis\u00e9. L\u2019optimisation de cette phase d\u2019adaptation reste encore n\u00e9cessaire pour assurer l\u2019efficacit\u00e9 et la rentabilit\u00e9 du proc\u00e9d\u00e9 industriel.<\/p><p>Ici, nous avons \u00e9tudi\u00e9 le r\u00f4le du facteur de transcription XylR dans la dur\u00e9e des phases de latence lorsque la bact\u00e9rie <em>E. coli <\/em>doit s&#8217;adapter de la croissance \u00e0 base de glucose \u00e0 celle de xylose. Tout d&#8217;abord, divers temps de latence ont \u00e9t\u00e9 observ\u00e9s lorsque diff\u00e9rentes souches d&#8217;<em>E. coli <\/em>passent d\u2019une croissance sur glucose \u00e0 une croissance sur xylose. Ces temps de latence se sont av\u00e9r\u00e9s \u00eatre contr\u00f4l\u00e9s par la disponibilit\u00e9 de XylR dans les cellules. Le titrage de XylR provoque de longs temps de latence r\u00e9sultant d\u2019un ph\u00e9nom\u00e8ne d\u2019h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 ph\u00e9notypique lors du passage du glucose au xylose, avec une sous-population incapable de reprendre une croissance exponentielle, tandis que l&#8217;autre sous-population augmente de fa\u00e7on exponentielle en consommant le xylose. Un mod\u00e8le stochastique a ensuite \u00e9t\u00e9 construit sur la base de l&#8217;hypoth\u00e8se que la disponibilit\u00e9 de XylR influence la probabilit\u00e9 des cellules individuelles de passer \u00e0 la croissance du xylose. Le mod\u00e8le a \u00e9t\u00e9 utilis\u00e9 pour montrer comment XylR se comporte comme un commutateur mol\u00e9culaire d\u00e9terminant deux populations ph\u00e9notypiquement distinctes.<\/p><p>Ce travail montre que la longueur des phases de latence chez <em>E. coli<\/em> est contr\u00f4lable et renforce le r\u00f4le des m\u00e9canismes stochastiques dans l&#8217;adaptation cellulaire, ouvrant la voie \u00e0 de nouvelles strat\u00e9gies pour une meilleure utilisation des sources de carbone durables en bio\u00e9conomie.<\/p><p>Ces recherches ont fait l\u2019objet d\u2019un article dans la revue internationale \u00e0 comit\u00e9 de lecture <em>mBio<\/em> (doi.org\/10.1128\/mBio.02938-20).<\/p><p>Elles ont \u00e9t\u00e9 initi\u00e9es par un financement 3BCAR (projet HME) et ont aussi servi de base au montage d\u2019un projet ANR (JANUS ; ANR-19-CE43-0004).<\/p><p><strong>R\u00e9f\u00e9rences bibliographiques<\/strong> :<\/p><p>Barthe, M., Tchouanti ,J., Gomes, P.H., Bideaux, C., Lestrade, D., Graham, C., Steyer, J.P., Meleard, S., Harmand, J., Gorret, N., Cocaign-Bousquet, M., &amp; Enjalbert, B.
