class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Cet axe concerne <strong>la compr\u00e9hension de l&#8217;h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 cellulaire pour ma\u00eetriser la robustesse des souches microbiennes en condition de bioproduction intensive<\/strong>.<\/p><p>Les recherches d\u00e9velopp\u00e9es dans cet axe portent sur la caract\u00e9risation et la quantification des stabilit\u00e9s g\u00e9n\u00e9tique (g\u00e9nome et\/ou plasmide) et non-g\u00e9n\u00e9tique (\u00e9pig\u00e9n\u00e9tique, expression de g\u00e8nes, m\u00e9tabolisme) ainsi qu\u2019aux variabilit\u00e9s des \u00e9tats physiologiques (capacit\u00e9 d\u2019accumulation, viabilit\u00e9, int\u00e9grit\u00e9 membranaire, etc) dans le cadre de proc\u00e9d\u00e9s biotechnologiques. L\u2019objectif est de quantifier les dynamiques de la r\u00e9ponse microbienne et de la robustesse des souches (\u00e0 l\u2019\u00e9chelle de la population globale et des sous-populations) \u00e0 des fluctuations d\u2019environnement, des conditions de stress et des conditions de production intensive afin d\u2019intensifier \/ optimiser les bioproc\u00e9d\u00e9s et leur changement d\u2019\u00e9chelle. De plus, l\u2019\u00e9tude des cons\u00e9quences de la variabilit\u00e9 d\u2019origine mol\u00e9culaire sur l\u2019h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 ph\u00e9notypique intercellulaire est \u00e9tudi\u00e9e. Les travaux de cet axe sont r\u00e9alis\u00e9s dans la perspective de pouvoir utiliser l\u2019ensemble de ces informations pour r\u00e9tro-ing\u00e9ni\u00e9rer des souches microbiennes et am\u00e9liorer leur comportement et robustesse en condition de bioproduction mais aussi, par la prise en compte de la distribution des sous-populations au cours du bioproc\u00e9d\u00e9, de mettre en place des boucles de r\u00e9trocontr\u00f4le sur les strat\u00e9gies de conduite des bior\u00e9acteurs. Il s\u2019agit notamment de caract\u00e9riser finement les cons\u00e9quences des conditions de productions intensive (h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9s dans les bior\u00e9acteurs, nombre de g\u00e9n\u00e9rations et densit\u00e9 cellulaire \u00e9lev\u00e9e) sur l\u2019h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 intercellulaire, afin de d\u00e9velopper des outils de conversion of CO<sub>2 <\/sub>by smart autotrophic biorefineries<\/em> (H2020-MSCA-ITN ConCO<sub>2<\/sub>rde)<\/p><p>L&#8217;ITN CONCO<sub>2<\/sub>RDE EJD est un r\u00e9seau de formation par la recherche de 11 doctorants sur des projets de recherche de pointe alliant biologie synth\u00e9tique, ing\u00e9nierie m\u00e9tabolique et g\u00e9nie des bioproc\u00e9d\u00e9s pour la transformation microbienne du CO<sub>2<\/sub>\/H<sub>2<\/sub>\/air en mol\u00e9cules d&#8217;int\u00e9r\u00eat diverses. Pour ce faire, les niveaux de production doivent \u00eatre adapt\u00e9s en permanence pour contrebalancer les d\u00e9rives g\u00e9n\u00e9tiques\/m\u00e9taboliques ou les changements de population microbienne. Le projet a ainsi pour objectif :<\/p><ol><li>de concevoir une r\u00e9gulation g\u00e9n\u00e9tique autonome, assist\u00e9e ou contr\u00f4l\u00e9e par l&#8217;utilisateur, du m\u00e9tabolisme microbien dans les bior\u00e9acteurs,<\/li><li>de construire divers types de capteurs capables de mesurer les param\u00e8tres des cellules ou des esp\u00e8ces et<\/li><li>de contr\u00f4ler, en temps r\u00e9el, les orientations m\u00e9taboliques des microorganismes \u00e0 l&#8217;aide de d\u00e9clencheurs externes utilisables dans les bior\u00e9acteurs industriels.<\/li><\/ol><p>Dans le cadre de ce projet, l\u2019\u00e9quipe est impliqu\u00e9e plus particuli\u00e8rement dans le WP1 avec pour objectif de d\u00e9velopper une approche de caract\u00e9risation <em>in situ<\/em> qualitative et quantitative des \u00e9tats physiologiques des cellules par cytom\u00e9trie en flux coupl\u00e9e \u00e0 une boucle de r\u00e9trocontr\u00f4le sur un actionneur du bior\u00e9acteur afin de mieux prendre en compte le comportement d\u2019une population microbienne et pouvoir ajuster en cons\u00e9quence les param\u00e8tres de conduites du fermenteur. Le microorganisme, par une mesure de son comportement, deviendrait en quelque sorte le pilote du bior\u00e9acteur. Dans ce contexte, ce projet vise \u00e0 d\u00e9velopper des strat\u00e9gies innovantes pour am\u00e9liorer l&#8217;\u00e9tape de fermentation de la production de biocarburants dans la bioraffinerie, d\u00e9velopper de nouvelles alternatives pour la valorisation des effluents gazeux et proposer de nouvelles strat\u00e9gies pour la valorisation de la lignine. Dans le cadre de ce projet, l\u2019\u00e9quipe est particuli\u00e8rement impliqu\u00e9e dans le WP1, T\u00e2che 2.1, \u00e0 savoir la valorisation des flux gazeux (CO<sub>2<\/sub>\/H<sub>2<\/sub>) en alcanes par des souches de <em> necator<\/em> g\u00e9n\u00e9tiquement modifi\u00e9es. Afin de r\u00e9pondre aux grands enjeux soci\u00e9taux de lutte contre le r\u00e9chauffement climatique, l\u2019\u00e9puisement des ressources fossiles, la mobilisation des surfaces agricoles, l\u2019accroissement rapide de la population, il est devenu essentiel de rechercher des solutions de diversification des sources d\u2019approvisionnement. \u00c0 cet \u00e9gard, les levures ol\u00e9agineuses sont des candidats prometteurs, de par leur capacit\u00e9 \u00e0 cro\u00eetre rapidement en bior\u00e9acteur, ainsi que pour leur capacit\u00e9 \u00e0 moduler leur m\u00e9tabolisme et leur composition macroscopique en fonction des conditions de croissance. <em>Yarrowia lipolytica<\/em> connue pour ses capacit\u00e9s naturelles d\u2019accumulation de lipides, et d\u2019utilisation de sources de carbone diverses a \u00e9t\u00e9 choisie. Cependant, en r\u00e9ponse aux fluctuations environnementales, <em> lipolytica<\/em> d\u00e9clenche des transitions dimorphiques g\u00e9n\u00e9rant des difficult\u00e9s de mise en \u0153uvre des bioproc\u00e9d\u00e9s (perte de rendement, limitation des transferts, impact sur la rh\u00e9ologie des milieux). Dans ce contexte, le projet porte sur l\u2019\u00e9tude de l\u2019impact de la filamentation sur les capacit\u00e9s de production de lipides (titre et composition) et r\u00e9ciproquement. Diff\u00e9rentes conditions environnementales et nutritionnelles sont \u00e9tudi\u00e9es afin d\u2019identifier les m\u00e9canismes de r\u00e9gulation mis en \u0153uvre.
