microorganismes comme usine cellulaire pour valoriser des ressources renouvelables en bioproduits. Nous cr\u00e9ons des usines cellulaires microbiennes en dotant divers micro-organismes d\u2019int\u00e9r\u00eat industriel (<em>E. coli, B. methanolicus, Y. lipolytica<\/em>) de nouvelles fonctions qui n\u2019existent pas n\u00e9cessairement dans la nature. Nous m\u00e9langeons et associons des enzymes m\u00e9taboliques ou des prot\u00e9ines r\u00e9gulatrices de diff\u00e9rentes esp\u00e8ces afin d\u2019\u00e9largir le portefeuille de substrats et de produits de ces micro-organismes. L\u2019analyse de la r\u00e9ponse int\u00e9gr\u00e9e du microorganisme \u00e0 ces nouvelles fonctionnalit\u00e9s m\u00e9taboliques ou r\u00e9gulatrices permet de comprendre leur fonctionnement et donc data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t\t\t\t\t\t<p>Dans ce projet, nous proposons de remplacer les mati\u00e8res premi\u00e8res entrant en concurrence avec l&rsquo;alimentation, actuellement utilis\u00e9es en biotechnologie industrielle, par le m\u00e9thanol, afin de promouvoir une bio\u00e9conomie circulaire et durable. Le m\u00e9thanol constitue une alternative prometteuse en tant que source de carbone renouvelable, puisqu\u2019il peut \u00eatre synth\u00e9tis\u00e9 \u00e0 partir de CO\u2082, contribuant ainsi aux efforts de lutte contre le changement climatique. Notre travail consiste \u00e0 reprogrammer l\u2019architecture m\u00e9tabolique d\u2019<em>E. coli <\/em>pour lui permettre d\u2019assimiler du m\u00e9thanol en tant que source principale de carbone et d\u2019\u00e9nergie. La reprogrammation du r\u00e9gime alimentaire d\u2019<em>E coli <\/em>pour l\u2019assimilation d\u2019une nouvelle source de carbone tel que le m\u00e9thanol est une t\u00e2che difficile. Elle ne n\u00e9cessite pas seulement d\u2019\u00e9quiper <em>E. coli<\/em> avec une voie m\u00e9thylotrophique synth\u00e9tique. Il est \u00e9galement indispensable d\u2019int\u00e9grer cette nouvelle fonctionnalit\u00e9 m\u00e9tabolique avec les processus cellulaires globaux. C\u2019est ce que nous proposons de faire dans ce projet : (i) en construisant un r\u00e9seau de r\u00e9gulation semi-synth\u00e9tique qui r\u00e9pond au m\u00e9thanol et active l\u2019ensemble des processus cellulaires natifs d\u2019 <em>E. coli\u00a0<\/em>; et (ii) en isolant la voie m\u00e9thylotrophique synth\u00e9tique au sein de microcompartiments bact\u00e9riens (BMCs). Dans la premi\u00e8re \u00e9tape, le CO\u2082 est converti en ac\u00e9tate par des communaut\u00e9s microbiennes ana\u00e9robies, dans des conditions contr\u00f4l\u00e9es en bior\u00e9acteurs. Dans la seconde \u00e9tape, cet ac\u00e9tate est utilis\u00e9 comme source de carbone par des souches d\u2019<em>E. coli <\/em>modifi\u00e9es g\u00e9n\u00e9tiquement pour produire diverses biomol\u00e9cules d\u2019int\u00e9r\u00eat industriel. Nous combinons l\u2019ing\u00e9nierie enzymatique (en collaboration avec l\u2019\u00e9quipe CIMES) et l\u2019ing\u00e9nierie m\u00e9tabolique pour adapter la composition en acides gras en fonction des compos\u00e9s d\u2019int\u00e9r\u00eat.
