th\u00e8me, l\u2019objectif est de comprendre comment les microorganismes (en particulier <em>E. coli, B. methanolicus, P. pastoris<\/em>, <em>C. tetani<\/em>) d\u00e9veloppent des r\u00e9ponses m\u00e9taboliques int\u00e9gr\u00e9es aux changements environnementaux et\/ou g\u00e9n\u00e9tiques. Classiquement, une telle r\u00e9ponse implique des m\u00e9canismes complexes et notre objectif est de comprendre pr\u00e9cis\u00e9ment comment les r\u00e9seaux m\u00e9taboliques et de r\u00e9gulations (naturels ou synth\u00e9tiques !) coop\u00e8rent dans la r\u00e9ponse int\u00e9gr\u00e9e de la cellule. Nous exploitons ensuite ces connaissances pour am\u00e9liorer la conception de microorganismes utiles \u00e0 l\u2019industrie\u00a0(<a l\u2019ac\u00e9tate mais il peut \u00e9galement modifier les prot\u00e9ines par phosphorylation ou ac\u00e9tylation. L\u2019ac\u00e9tylation des prot\u00e9ines chez les bact\u00e9ries se produit par l\u2019action d\u2019une ac\u00e9tyltransf\u00e9rase utilisant l\u2019ac\u00e9tyl-CoA ou de mani\u00e8re non enzymatique par l\u2019AcP. Une grande partie des enzymes du m\u00e9tabolisme central peuvent \u00eatre ac\u00e9tyl\u00e9es, ce qui sugg\u00e8re un r\u00f4le de l\u2019ac\u00e9tylation dans la r\u00e9gulation du m\u00e9tabolisme. Cette vision a commenc\u00e9 \u00e0 \u00e9voluer avec la d\u00e9couverte des microcompartiments bact\u00e9riens (BMCs). Les BMCs sont des structures prot\u00e9iques hautement organis\u00e9es, constitu\u00e9es d\u2019une enveloppe semi-perm\u00e9able qui encapsule des enzymes sp\u00e9cifiques. L\u2019ac\u00e9tate d\u00e9tourne le carbone du m\u00e9tabolisme cellulaire et il est toxique pour les cellules, diminuant ainsi la productivit\u00e9 des bioproc\u00e9d\u00e9s. L\u2019origine, le contr\u00f4le et la r\u00e9gulation de la production d\u2019ac\u00e9tate restent \u00e0 clarifier. Dans ce projet, nous abordons des questions fondamentales et appliqu\u00e9es sur le r\u00f4le et la r\u00e9gulation de la production d\u2019ac\u00e9tate chez <em>E. coli<\/em>, dans le but de proposer des strat\u00e9gies pour am\u00e9liorer la productivit\u00e9 des bioproc\u00e9d\u00e9s bas\u00e9s sur <em>E. physiologie de la bact\u00e9rie <em>Clostridium tetani<\/em> aux niveaux macroscopique, m\u00e9tabolique et prot\u00e9omique afin de d\u00e9crypter les m\u00e9canismes d\u2019induction de la synth\u00e8se de la toxine t\u00e9tanique.
